Anotace: Healthy VF mucosa is composed of two major cell types, non-keratinized stratified squamous epithelium and fibroblasts that reside in the underlying lamina propria. Although dysfunction of the epithelium may play a significant pathogenic role in VF diseases, studies at the genetic and molecular level using primary VF epithelial cells or models of human VF mucosa have been significantly limited by the availability of relevant tissue types derived from biopsies or cadaveric sources, poor growth, and heterogeneity of primary VF epithelial cells.  To overcome these challenges, we developed a three-dimensional model of human VF mucosa using human induced pluripotent stem cell-derived (hiPSC-derived) VF epithelium and primary VF fibroblasts embedded in collagen matrix that represents the lamina propria. This 3D system uses an unlimited source of VF epithelial cells which differentiated from hiPSCs mimicking their embryonic development. We further provided functional validation of engineered VF mucosae and exposed them to distinct environmental insults such as cigarette smoke and electronic cigarette vaping extracts to model hyperkeratotic mucosal changes, epithelial lipid-mediated VF injury and mucosal inflammation. Our findings suggest that the hiPSC-derived model of human VF mucosa can provide valuable information about VF mucosal cell behavior in response to environmental challenges and holds great promise for the investigation of toxicity and mechanisms of tobacco or vaping-related cellular injuries/pathologies in human VF stratified epithelium.
